Abstract:
This literature review focuses on patients with congenital leptin deficiency who had never been exposed to endogenous or exogenous leptin and its effect on energy intake, body weight, body fat mass percentage, genetic leptin deficiency phenotype, serum levels of leptin, metabolic and hormonal parameters ( lipids profile, blood sugar parameters and gonadotropic hormones). Most available data on leptin replacement therapy were cross over studies. The aim of this review is to identify patients with genetic leptin deficiency, its phenotypes and to explore leptin replacement therapy among individuals who are only naïve for leptin. The review demonstrated that leptin replacement therapy has powerful effects and its phenotypes are almost reversible, and that affected cases are under estimated.
